This experiment was conducted to determine the most effective medium composition and optimal concentrations of plant growth regulators for rapid mass propagation and seedling growth of Sarcanthus scolopendrifollus, epiphytic rare and dangered orchid species in Korea. The effective medium for shoot differentiation and seedling growth of the orchid was Murashige and Skoog´s basal medium supplemented with 5% sucrose and 0.7% agar, and the medium was adjusted to pH 4.0. Number of shoot and fresh weight were enhanced in the medium added with 500 ㎎·L^(-1) activated charcoal, but root growth was decreased. Seedlings were died in the medium treated with 1.0 ㎎·L^(-1) NAA. However, shoot number, shoot length, leaf number, root number, root length and fresh weight were markedly increased in MS medium containing 10.0 ㎎·L^(-1) BA with 0.1 ㎎·L^(-1) NAA.
